重新定位Disulfiram引發銅依賴性之細胞銅死亡以對抗大腸直腸癌之新用途 (Repurposing Disulfiram as anti-cancer drug via induction cupper-dependent cuproptosis)

為了找尋針對屬於最易復發且具有最差之整體生存率的CMS4大腸直腸癌(CRC)分型之藥物,我們先前已利用大數據資料庫進行分析並發掘Disulfiram(DSF)為一潛力藥物。我們的近期研究證實DSF藉其做為銅離子載體,可有效抑制大腸直腸癌細胞之增殖。DSF合併銅離子之處理,可誘導一有別於凋亡之銅死亡機制,其可藉促進DLAT蛋白質之聚化而降低粒線體的代謝活性;此現象在三維的CRC腫瘤球體模式亦可看到相當的抑制現象,說明DSF未來可做為大腸直腸癌治療之新選擇!
